Dylan Summers-Jones’ ninth goal of the season via a 40th minute penalty was enough to clinch this tie for Hotspur.
The Holy Islanders completed a quick home double over Borough, having beaten them 2-0 in the league at the New Stadium a week earlier.
In Saturday’s League Cup clash, Aron Hughes Jones set up Kane Roberts for a 13th minute opener before Jack Williams levelled on 29.
Summers-Jones’ penalty also saw a red card for Conwy’s Ryan Vaughan result from the same incident.
Llanrhaeadr-ym-Mochnant 4 Cefn Albion 2
For the second weekend running, Albion left Llanrhaeadr as losers, Saturday’s 4-2 reverse coming in the League Cup.
Two Billy Clarke goals on 15 and 24 minutes put the hosts in command, but Callum Mannion and Torne Samuels brought Cefn level before Clarke completed his hat-trick just prior to the interval.
Albion had Jamie Foulkes and new interim manager Sean Jones red-carded in the second half before William Roberts-Morris sealed it for Llan with a 95th minute fourth goal.
Llanuwchllyn 1 Builth Wells 1 (Builth won 4-2 on penalties)
Builth progressed 4-2 on penalties after the tie itself had ended 1-1.
Meilir Williams nudged Llanuwchllyn in front on 10 minutes with his 18th goal of the season, but Tommy Rowlands made it one-apiece before the interval.
In a goalless second half, Warren Aykroyd-Duckett of Llanuwchllyn was sent off for a second yellow on 90 minutes.
Llay Welfare 0 Bethesda Athletic 1
While their league form has been disappointing, Bethesda’s cup results continue to impress.
Having already won at Llangefni Town in this competition, Mark Hughes’ men celebrated an equally great victory at Llay on Saturday.
Both managers were red-carded – Steven Halliwell for Llay and Pesda’s Hughes.
Osian Hughes notched a 68th minute decider for Athletic, who had Cian Williams receive a straight red in added time.
Losing manager Halliwell said: “Losing one-nil in a game that we absolutely dominated from start to finish, and to miss the amount of chances that we did, is really hard to take.
“Bethesda defended brilliantly all game, and caught us with a sucker punch that we knew was coming, because we kept missing these chances. Really disappointed to be out of the cup, especially in this fashion, but congratulations to Bethesda on the victory.”

A welcome win for Rhos, with Scott Reeves, a Jack Goulborn own goal, Aled Parry strike and Tom Smith double seeing them into the next round.
CPD Y Felinheli 1 Nantlle Vale 3

After four defeats on the bounce, Nantlle Vale deservedly returned to winning ways at Cae Seilo.
Cian Pritchard and Aled Williams both scored late in the first half after captain Aled Griffith had handed Felinheli a 19th minute lead.
Despite having assistant-manager Carl Vaughan Jones red-carded on 73 minutes, Vale clinched it when Jamie Jones made it 3-1 in the 94th.
Felin had Morgan Williams dismissed for two yellows with 87 on the clock.
