Barker worked wonders to win the ball on the left, race into the box and square to Ava Suckley whose shot was blocked but ex-Blackburn and Bolton midfielder Knox was following up to drill powerfully past keeper Mackenzie Haydn-Jones.
Both teams went close on other occasions, Caitlin Chapman and Kay Baker rattling the crossbar for TNS, while Mariam Mahmood was denied by a great Haydn-Jones save before striking the upright for the hosts.
Wrexham remain second in the table, level on points with Cardiff City, who head the standings on goal difference.
